Effect of oxytocin on gastric ischemia-reperfusion injury in rats

摘要: The effect of peripherally administered oxytocin (OT) on gastric ischemia-reperfusion injury (GI-RI) and its possible mechanism were investigated. The Sprague-Dawley (SD) rats were randomly divided into different treatment groups ( = 6). The animal GI-RI model was established by clamping the celiac artery for 30 min to induce ischemia and then released to allow reperfusion for 1 h, and the degree of GI-RI was assessed by scoring the gastric mucosal damage index (GMDI), the gastric fluid output, gastric fluid output, gastric acidity were measured and the surgical preparations of vagotomy and sympathectomy were used to investigate the possible mechanism of OT on GI-RI. The results were as follows. Compared with the control group (NS plus GI-R only, GMDI 121.33±10.40, = 6), the intra peritoneal (ip) administration of oxytocin (20, 100 μg/0.5 mL) obviously attenuated GI-RI (<0.05), GMDI were 82.33±14.26, 53.5±5.58 respectively ( = 6); the gastric fluid output and the gastric acidity (evaluated by pH) of the control group were (430.17±87.36) μL, 1.55±0.25 ( = 6), and those of the OT group were (102.45±48.00) μL, 2.65±0.40 ( = 6) res pectively; differences had statistical significance (<0.01). The effect of oxytocin was reversed by atosiban, a selective oxytocin receptor antagonist. The GMDI of the group given atosiban 10 min before OT was 138.17±24.06 ( = 6), which had no significant difference with the control group. Oxytocin further attenuated GI-RI after vagotomy and sympathectomy (GMDI 6.83±8.89, 29.67±5.54, = 6), compared with the GI-R group and the oxytocin group (<0.01). These results indicated that the oxytocin could significantly protect gastric mucosal against injury induced by ischemia-reperfusion, and the oxytocin receptor was involved. This effect of oxytocin may be mediated through the vagus and sympathetic nerve, and then lead to the reduction of gastric juice output and the depression of gastric acidity.

Environmental pollution and DNA methylation: carcinogenesis, clinical significance, and practical applications

摘要:

Environmental pollution is one of the main causes of human cancer. Exposures to environmental carcinogens result in genetic and epigenetic alterations which induce cell transformation. Epigenetic changes caused by environmental pollution play important roles in the development and progression of environmental pollution-related cancers. Studies on DNA methylation are among the earliest and most conducted epigenetic research linked to cancer. In this review, the roles of DNA methylation in carcinogenesis and their significance in clinical medicine were summarized, and the effects of environmental pollutants, particularly air pollutants, on DNA methylation were introduced. Furthermore, prospective applications of DNA methylation to environmental pollution detection and cancer prevention were discussed.

Prognostic significance of clinicopathologic parameters in gastrointestinal stromal tumors: a study of

摘要: The biological behavior of gastrointestinal stromal tumors (GISTs) are highly variable. To investigate the biological behavior of GIST, we collected 83 cases of gastric and 62 cases of small intestinal GIST from the Department of Pathology of the Chinese PLA General Hospital. The parameters include age, primary tumor location, tumor diameter, mitotic index, tumor necrosis, risk assessment, clinical stage and the exon 11 mutation. All these were analyzed in 105 cases along with the follow-up data and tested by log rank and COX hazard proportional model. We find that the average age of gastric GIST was 55.4 years. Of the 62 cases that were followed up, 17 cases had metastasis or recurrence and the 5-year survival rate was (66.51 ± 17.06)%. For the small intestinal GIST, the average age was 50.6 years and 43 cases were followed up. Of these, 22 cases had metastasis or recurrence and the 5-year survival rate was (61.76 ± 18.30)%. Small intestinal GIST was more frequently associated with metastasis and tumor relapse than gastric GIST ( = 6.131, = 0.013). For gastric GIST, patients younger than 50 years ( = 0.046), the advanced clinical stage ( = 0.0001), the large tumor diameter ( = 0.0001), a high mitotic index ( = 0.0001), necrosis ( = 0.0001) and a high risk grade ( = 0.004) were all correlated with a lower survival rate. The COX hazard proportional model revealed that advanced clinical stage ( = 0.001), large tumor size ( = 0.001), a high mitotic index ( = 0.002) and the high risk grade ( = 0.018) indicated a poorer prognosis in gastric GIST. For small intestinal GIST, necrosis ( = 0.036) and advanced clinical stage ( = 0.010) were associated with lower survival rates and the clinical stage was shown to be an independent prognostic indicator. A total of 25 cases harbored mutations in exon 11. The frequency of mutation was 32% and 22.5% for gastric and small intestinal GIST, respectively. In gastric GIST, the mutated was predominant in patients older than 50 years of age. But in the small intestinal GIST, the mutated was predominant in the age group of 40–49 years. In conclusion, for gastric GIST, clinical stage, tumor size, mitotic index, and risk grade are the prognostic indicators. For small intestinal GIST, necrosis and clinical stage are the prognostic indicators. Small intestinal GIST are more aggressive than gastric GIST. The occurrence of mutation may correlate with the age of patients.

Change and significance of T-cell subsets and TNF-α in patients with advanced malignant obstructive jaundice

摘要: The aim of this article was to study the influence of immunity function of advanced malignant obstructive jaundice (MOJ) treated by percutaneous transhepatic biliary external and internal drainage. Ninety-six cases of MOJ were divided into two groups according to the different ways of biliary drainage. Fifty-two external drainage tubes were placed in 41 cases of percutaneous transhepatic biliary external drainage group and 66 metal stents were placed in 55 cases of percutaneous transhepatic biliary internal drainage group. Liver function, serum TNF- and cellular function were examined one day before operation and one week after operation and liver function was re-examined two weeks after operation, in order to observe the change and analyze the asso ciation among them and compare with the control group. All patients conditions were improved after operation. In the percutaneous transhepatic biliary external and internal drainage groups, the total level of bilirubin decreased from (343.54±105.56) μ mol/L and (321.19±110.50) μ mol/L to (290.56±103.46) μ mol/L and (283.72±104.95) μ mol/L after operation respectively, which were significantly lower than pre-operation (<0.05), but there was no significant difference between the two groups (>0.05). Serum alanine aminotransferase (ALT) of all patients one week after operation was significantly lower than that before operation. TNF- in percutaneous transhepatic biliary external and internal groups decreased from (108.58±19.95) pg/mL, (109.98±16.24) pg/mL of pre-operation to (104.32±19.59) pg/mL, (83.92±13.43) pg/mL of post-operation respectively, there was notable improvement (<0.01) in internal drainage group after operation. Patients serum CD4, CD3 and CD4/CD8 were notably increased, but CD8 was notably decreased (<0.05). There was no difference in external drainage group (>0.05). There was a significant difference between the two groups. Serum TNF- and ALT had positive correlation. Percutaneous transhepatic biliary internal or external drainage was an effective and important method to treat MOJ. Patients immune function was weak when they suffered MOJ, but body s cellular immune function can be notably improved after internal biliary drainage.

Factor analysis for the statistical modeling of earthquake-induced landslides

摘要: Earthquake-induced landslides are difficult to assess and predict owing to the inherent unpredictability of earthquakes. In most existing studies, the landslide potential is statistically assessed by collecting and analyzing the data of historical landslide events and earthquake observation records. Unlike rainfall-induced landslides, earthquake-induced landslides cannot be predicted in advance using real-time monitoring systems, and the development of the models for these landslides should instead depend on early earthquake warnings and estimations. Hence, in this study, factor analysis was performed and the frequency distribution method was employed to investigate the potential risk of the landslides caused by earthquakes. Factors such as the slope gradient, lithology (geology), aspect, and elevation were selected and classified as influential factors to facilitate the construction of a landslide database for the area of study.

Analysis on the distinguishing features of traditional Chinese therapeutics and related statistical issues

摘要: Traditional Chinese medicine (TCM) is one of the rarely existing ancient traditional medicines that hold systematic theories as well as preventative and therapeutic methods for diseases in practice. From the 1950s, such research methods as mathematics, statistics, and data mining (DM) have been gradually introduced to TCM studies, making it more scientific. Meanwhile, the distinct features of TCM theories and diagnostic-model have constantly challenged the methodology of statistics. This paper introduces the following scientific features of traditional Chinese therapeutics: 1) its goal is to balance the functions and conditions of human body; 2) it emphasizes on holism and individualization; 3) it stresses the longitudinal regulation and evaluation mode, which is a circle of syndrome diagnosis, treatment and evaluation; 4) the interventions of TCM are abundant, compound and natural; and 5) humanistic thought is everywhere. Some statistical problems are raised based on these features. First, complex statistical methods that can analyze subjective indexes and latent variables, multidimensional and multistage data, non-equilibrium designed studies, and longitudinal data are required. Second, comprehensive evaluation on multiple-target mechanism has been brought in by combination treatment. Third, there is a need to analyze how humanity and related cultural factors may influence the effect of interventions. Thus, promoting implemented studies of statistics as well as carrying out the TCM scientific propositions have become the common expectations of both TCM and modern medicine.

It is well documented that patients can show significant heterogeneous responses to treatments so the best treatment strategies may require adaptation over individuals and time. Recently, a number of new statistical methods have been developed to tackle the important problem of estimating personalized treatment rules using single-stage or multiple-stage clinical data. In this paper, we provide an overview of these methods and list a number of challenges.

Statistical considerations for genomic selection

摘要: Genomic selection is becoming increasingly important in animal and plant breeding, and is attracting greater attention for human disease risk prediction. This review covers the most commonly used statistical methods and some extensions of them, i.e., ridge regression and genomic best linear unbiased prediction, Bayesian alphabet, and least absolute shrinkage and selection operator. Then it discusses the measurement of the performance of genomic selection and factors affecting the prediction of performance. Among the measurements of prediction performance, the most important and commonly used measurement is prediction accuracy. In simulation studies where true breeding values are available, accuracy of genomic estimated breeding value can be calculated directly. In real or industrial data studies, either training-testing approach or -fold cross-validation is commonly employed to validate methods. Factors influencing the accuracy of genomic selection include linkage disequilibrium between markers and quantitative trait loci, genetic architecture of the trait, and size and composition of the training population. Genomic selection has been implemented in the breeding programs of dairy cattle, beef cattle, pigs and poultry. Genomic selection in other species has also been intensively researched, and is likely to be implemented in the near future.

Statistical process control with intelligence using fuzzy ART neural networks

摘要: With the automation development of manufacturing processes, artificial intelligence technology has been gradually employed to increase the automation and intelligence degree in quality control using statistical process control (SPC) method. In this paper, an SPC method based on a fuzzy adaptive resonance theory (ART) neural network is presented. The fuzzy ART neural network is applied to recognize the special disturbance of the manufacturing processes based on the classification on the histograms, which shows that the fuzzy ART neural network can adaptively learn the features of the histograms of the quality parameters in manufacturing processes. As a result, the special disturbance can be automatically detected when a feature of the special disturbance starts to appear in the histograms. At the same time, combined with spectrum analysis of the autoregressive model of quality parameters, the fuzzy ART neural network can also be utilized to adaptively detect the abnormal patterns in the control chart.

Assessment of temporal and spatial variations in water quality using multivariate statistical methods

摘要: This study evaluated the temporal and spatial variations of water quality data sets for the Xin'anjiang River through the use of multivariate statistical techniques, including cluster analysis (CA), discriminant analysis (DA), correlation analysis, and principal component analysis (PCA). The water samples, measured by ten parameters, were collected every month for three years (2008–2010) from eight sampling stations located along the river. The hierarchical CA classified the 12 months into three periods (First, Second and Third Period) and the eight sampling sites into three groups (Groups 1, 2 and 3) based on seasonal differences and various pollution levels caused by physicochemical properties and anthropogenic activities. DA identified three significant parameters (temperature, pH and ) to distinguish temporal groups with close to 76% correct assignment. The DA also discovered five parameters (temperature, electricity conductivity, total nitrogen, chemical oxygen demand and total phosphorus) for spatial variation analysis, with 80.56% correct assignment. The non–parametric correlation coefficient (Spearman R) explained the relationship between the water quality parameters and the basin characteristics, and the GIS made the results visual and direct. The PCA identified four PCs for Groups 1 and 2, and three PCs for Group 3. These PCs captured 68.94%, 67.48% and 70.35% of the total variance of Groups 1, 2 and 3, respectively. Although natural pollution affects the Xin'anjiang River, the main sources of pollution included agricultural activities, industrial waste, and domestic wastewater.

Insult of gastroesophageal reflux on airway: clinical significance of pharyngeal nozzle

摘要:

At the very time of global paying the highest attention to the worst insults of smoking as well as haze on the airway, everybody knows both are exogenous and noticeable. However, people mostly, including many medical personnel, do not know how badly the gastroesophageal reflux (GER) insults on our own airway. Symptoms of GER are commonly seen as heartburn and regurgitation, which can be mostly tolerated. However, when the up going gastric content reversely passes the esophagus and then the distal pharynx, where it appears a beak like stricture, serving as a nozzle, so as to produce numerous micro-particles and reach the oro-nasal cavity and also the airway causing allergic rhinitis and asthmatic attacks, even pulmonary parenchyma lesions. It will reduce life quality or even jeopardize life. The point that the endogenous insult appears in the respiratory system, but originates from the digestive tract is not well known and often undiagnosed and not correctly treated. The GER induced airway challenge is a treatable and preventive entity, as soon as a diagnosis is made, a good relief could be expected by means of life style adjustment, medicine, or fixation of the patulous cardia through radiofrequency or fundoplication. The author Dr. Zhonggao Wang had suffered it for long and symptoms disappeared for 8 years after anti-reflux surgery. Here is a presentation of Dr. Zhonggao Wang and his team’s work and would call attention to the public so as to recognize this relatively unknown entity — a treatable condition occurring from human itself, but not from outside surroundings as smoking or haze does.

Scientific significance of ancient maps of Yellow River and Grand Canal for water conservancy in China

摘要: Based on the study of ancient maps preserved in China and abroad, the systematic nature and practical meaning of the maps of the Yellow River and Grand Canal is demonstrated. It is pointed out that the ancient maps not only record the spatial information of the established water conservancy engineering for river harnessing but also the management systems of the rivers in history. Besides, the maps provide abundant information on nature, humanity, and geography and possess high value in academic research and art appreciation.

Clinical significance of human papilloma virus infection in the cervical lesions

摘要: Cervical lesions have been regarded as the common and frequently occurring diseases in China. Recently, the morbidity and youth tendency of cervical cancer have gradually increased. Cervical cancer, related with human papilloma virus (HPV) infection, has been one of the severest diseases threatening health and life of women, and is an infectious disease. The universality of HPV infection in the reproductive tract should not be ignored. The well-known risk factors of HPV infection in cervical lesions consist of high-risk sexual behaviors, immunosuppression, age, contraceptive methods, the concurrent infection of other sexually transmitted diseases, etc. The variation of cervical lesions induced by HPV infection is involved in the continuous pathological process, including the subclinical, latent, and persistent infection of high risk (HR)-HPV, chronic cervicitis with abnormal results of cytological examination, cervical intraepithelial neoplasia (CIN), and cervical cancer. The outcome of patients with HPV infection is influenced by many factors, such as HPV subtype dominance, persistent HPV infection, HPV loading dose, and multiple HPV infection. Controlling HR-HPV persistent infection should be an important strategy for reducing cervical lesions.

Increasing significance of advanced physical/chemical processes in the development and application of

摘要: The awareness of the problem of the scarcity of water of high quality has strongly changed the approach of wastewater treatment. Currently, there is an increasing need for the beneficial reuse of treated wastewater and to recover valuable products and energy from the wastewater. Because microbiological treatment methods are, only to a limited part, able to satisfy these needs, the role and significance of physical/chemical processes in wastewater treatment are gaining more and more interest. The specific future role and aim of the various physical/chemical treatment processes can be categorized in five groups: improvement of the performance of microbiological treatment processes, achievement of the high quality required for reuse of the effluent, recovery of valuable components and energy from the wastewater for beneficial reuse, desalination of brackish water and seawater, and treatment of concentrated liquid or solid waste residues produced in a wastewater treatment process. Development of more environmentally sustainable wastewater treatment chains in which physical/chemical processes play a crucial role, also requires application of process control and modeling strategies. This is briefly introduced by the elaboration of treatment scenarios for three specific wastewaters.

Application of statistical design for the production of inulinase by

摘要: A Plackett-Burman design was employed for screening 18 nutrient components for the production of inulinase using sp. and pressmud as the substrate via solid-state fermentation (SSF). From the experiments, three nutrients viz. yeast extract, FeSO ·7H O, and NH NO were found to be the most significant components. Hence these three components were selected and optimized using Response Surface Methodology (RSM). The optimum conditions are: yeast extract 0.00274 g/gds, FeSO ·7H O 0.00011 g/gds and NH NO 0.00772 g/gds. The effect of the substrate concentration and initial moisture content were also studied. A substrate concentration of 12 g and an initial moisture content of 65% are optimum for the maximum production of inulinase (89 U/gds).
